The Bears swept Stirling Albion aside in the last round and will take on lower-league opposition yet again.
Jon McLaughlin will start in goal and will come in for Allan McGregor after the veteran nailed down his spot in recent months.
There will be three changes at the back, with Borna Barisic, Mateusz Zukowski and Jack Simpson all coming in.
Connor Goldson will keep his spot as usual, but James Tavernier, Leon Balogun and Calvin Bassey will get much-needed rest.
James Sands will anchor the midfield, after the American appeared to have fallen out of favour in recent weeks.
There will also be a spot for Steven Davis, who will finally come back into the side after weeks of trouble to dictate play on what could be a tough evening.
Aaron Ramsey will get his first start and earn a full debut, too, and it will be brilliant to see the Welsh international start from the first whistle.
Amad Diallo and Fashion Sakala will start on the wings, with pure pace and skill set to be real weapons for Annan to try and deal with.
Finishing the line-up will be Kemar Roofe, with Cedric Itten wasting numerous chances last month.
